DISRUPTING CXCR2 MACROMOLECULAR COMPLEX PDZ-DOMAININTERACTIONS DURING INFLAMMATORY CHEMOTAXISbyMARCELLO P. CASTELVETEREAugust 2012Advisor: Dr. Chunying Li Major: Biochemistry and Molecular BiologyDegree: Master of ScienceNeutrophils are the body’s first responders to inflammation being the mostabundant white blood cell type in circulation and they quickly initiate an immuneresponse through chemokine signaling. Inflammatory chemokines signal via theirreceptor CXCR2, which initiates an inflammatory response, recruiting leukocytes tosites of inflammation. Chemokine signaling is important for proper host protection, yet uncontrolled activity is responsible for a variety of pathological conditions: includingrheumatoid arthritis, ischemia-reperfusion injury, arteriosclerosis, multiple sclerosis, psoriasis, inflammatory bowel disease, and allergic reactions. In this report I show a CXCR2 macromolecular signaling complex exists inneutrophils containing NHERF1 and PLCβ2. I also demonstrate a novel strategy ofcytosolically perturbing the CXCR2 PDZ-domain interaction of the macromolecular complex. This perturbation disrupts spatial sensation of a chemokine gradient, yet stillallows cells to mobilize actin and chemotax. Furthermore, I show CXCR2 PDZ-domainperturbation does not disrupt migration through bacterial derived chemoattractantreceptors.

CXCR2 macromolecular complex in pancreatic cancer: a potential therapeutic target in tumor growth.
The signaling mediated by the chemokine receptor CXC chemokine receptor 2 (CXCR2) plays an important role in promoting the progression of many cancers, including pancreatic cancer, one of the most lethal human malignancies. CXCR2 possesses a consensus PSD-95/DlgA/ZO-1 (PDZ) motif at its carboxyl termini, which might interact with potential PDZ scaffold/adaptor proteins. We have previously repor...
متن کاملA critical role of CXCR2 PDZ-mediated interactions in endothelial progenitor cell homing and angiogenesis.
Bone marrow-derived endothelial progenitor cells (EPCs) contribute to neovessel formation in response to growth factors, cytokines, and chemokines. Chemokine receptor CXCR2 and its cognate ligands are reported to mediate EPC recruitment and angiogenesis. CXCR2 possesses a consensus PSD-95/DlgA/ZO-1 (PDZ) motif which has been reported to modulate cellular signaling and functions. Here we examine...
متن کاملStructural Insights into Neutrophilic Migration Revealed by the Crystal Structure of the Chemokine Receptor CXCR2 in Complex with the First PDZ Domain of NHERF1
Neutrophil plays an essential role in host defense against infection, but uncontrolled neutrophilic infiltration can cause inflammation and severe epithelial damage. We recently showed that CXCR2 formed a signaling complex with NHERF1 and PLC-2, and that the formation of this complex was required for intracellular calcium mobilization and neutrophilic transepithelial migration. To uncover the s...
متن کاملOpposing effects mediated by the chemokine receptor CXCR2 on myocardial ischemia-reperfusion injury: recruitment of potentially damaging neutrophils and direct myocardial protection.
BACKGROUND The timely reperfusion of ischemic myocardium limits infarction, but components of reperfusion, such as inflammation, may be injurious. The chemokine receptor CXCR2 mediates neutrophil chemotaxis. CXCR2 activation also inhibits hypoxia-induced death of isolated cardiac myocytes. This study assesses whether CXCR2 mediates protection in the intact heart and, if so, the magnitude of thi...

BACKGROUND Chemotaxis is essential for a number of physiological processes including leukocyte recruitment. Chemokines initiate intracellular signaling pathways necessary for chemotaxis through binding seven transmembrane G protein-couple receptors. Little is known about the proteins that interact with the intracellular domains of chemokine receptors to initiate cellular signaling upon ligand b...
